黄芩素对2型糖尿病大鼠糖脂代谢和肾功能的影响及其作用机制

摘    要:目的:探讨黄芩素对2型糖尿病(T2DM)大鼠糖脂代谢和肾功能的影响及其作用机制。方法:选择80只清洁级SD大鼠,其中20只喂食普通饲料8周,作为正常对照组,另60只喂食高糖高脂饲料8周,并腹腔注射链脲佐菌素(STZ)30 mg/kg诱导建立T2DM大鼠模型,建模成功后随机分为模型组、黄芩素高剂量组[160 mg/(kg·d)]和低剂量组[80 mg/(kg·d)]各20只,连续给药8周;分别于给药4周及8周时测定大鼠空腹血糖(FBG)、餐后2小时血糖(2 h PBG)及24小时尿蛋白含量,同时于给药8周后使用全自动分析仪测定血清总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)水平以及血清中血肌酐(SCr)、尿素氮(BUN)、尿肌酐(UCr),并计算肌酐清除率(Ccr);采用ELISA法测定血清血管内皮生长因子(VEGF)及血管紧张素Ⅱ(AngⅡ)的含量。结果:模型组大鼠血清FBG及2 h PBG水平均明显高于对照组,而在给药4周及8周时,低剂量组及高剂量组大鼠血清FBG及2 h PBG水平均较模型组明显降低,且高剂量组两指标水平降低程度均明显高于低剂量组,比较差异具有统计学意义(P0.05)。模型组大鼠TG、TC、LDL-C水平均明显高于对照组,而HDL-C水平明显低于对照组,比较差异具有统计学意义;给药8周后的低剂量组及高剂量组大鼠TG、TC、LDL-C及HDL-C水平均较对照组明显改善,且高剂量组改善程度明显高于低剂量组,比较差异具有统计学意义(P0.05)。模型组大鼠24小时尿蛋白、BUN、SCr、VEGF及AngⅡ水平均明显高于对照组,而Ccr明显低于对照组,在给药4周及8周后低剂量组及高剂量组大鼠24 h尿蛋白、BUN、SCr、Ccr、VEGF及AngⅡ水平均较模型组明显改善,而高剂量组改善程度明显高于低剂量组,比较差异具有统计学意义(P0.05)。结论:黄芩素可有效改善T2DM病情,延缓肾功能损害进展,值得临床推广应用。

Effects and Action Mechanism of Baicalein on Glucolipid Metabolism and Renal Function of the Rats with Type 2 Diabetes Mellitus
Abstract:Objective: To investigate the effects and action mechanism of baicalein on glucolipid metabolism and renal function of the rats with type 2 diabetes mellitus (T2DM). Methods: Eighty clean SD rats were selected, twenty rats as the normal control group were fed normal diets for eight weeks, other sixty were fed high-fat diets for eight weeks and accepted intraperitoneal injection of streptozotocin (STZ) 30mg /kg to found T2DM rat model, sixty rats were randomly divided into the model group, the high and low dosage groups of baicalein[160 mg/(kg·d) and 80 mg/(kg·d)], 20 rats each group, for eight weeks continuously; after continuous medication for four and eight weeks, FBG, 2 hPBG and urinary protein with in 24 hours were detected, the levels of TC,TG,LDL-C, HDL-C, SCr, BUN and UCr in serum were detected by automatic analyzer, and Ccr was calculated; the levels of VEGF and AngⅡwere detected by ELISA. Results: The levels of FBG and 2 hPBG in the model group were higher than these of the control group obviously, after medication for four and eight weeks, the levels of FBG and 2 hPBG of the rats in low and high dosages groups of baicalein decreased obviously compared with these of the model group, the dec rease of the indexes in the high dosage group were higher than these of the low dosage group obviously, there were statistical differences (P<0.05). The levels of TG, TC and LDL-C of rats in the model group were higher than these of the control group notably, but the level of HDL-C was lower than that of the control group, there were statistical differences; the levels of TG, TC, LDL-C and HDL-C of rats in low and high dosage groups after medication for eight weeks improved compared with these of the control group obviously, the improvements in the high dosage group were higher than these of the low dosage group remarkably, there were statistical differences (P<0.05). The levels of urinary protein with in 24 hours, BUN, SCr, VEGF and AngⅡof the rats in the model group were higher than these of the control group, the level of Ccr was lower than that of the control group, after medication for four and eight weeks, the levels of urinary protein with in 24 hours, BUN, SCr, Ccr, VEGF and AngⅡof the rats im-proved compared with these of the model group, the improvement in high dosage group was high than that of the
low dosage group, there were statistical differences (P<0.05). Conclusion: Baicalein can improve the condition of T2DM effectively and delay renal function injury progress, which is worthy of clinic application.
