卵巢癌耐药细胞系A2780/TS紫杉醇体外化疗后耐药相关基因的表达

摘    要:目的比较卵巢癌耐药与非耐药细胞系,紫杉醇体外化疗后部分耐药相关基因的表达差异。方法采用RT-PCR技术检测卵巢癌细胞系A2780与紫杉醇耐药细胞系A2780/TS部分耐药相关基因的表达差异。结果耐紫杉醇细胞株A2780/TS经紫杉醇处理后,细胞抑制率明显低于敏感细胞株A2780(P〈0.05);A2780/TS的耐药指数(RI)约为3.650;G1、G2、S期细胞比例无明显改变(P〉0.05);在检测的耐药相关基因中,A2780/TS中较其非耐药细胞中表达下降的有多药耐药蛋白(MDR-1)基因、P53,表达升高的有细胞色素P450181(CYP1B1)、凋亡抑制蛋白Survivin,无明显改变的有谷胱甘肽-S-转移酶(GST-Pi)、多药耐药相关蛋白(MRP-1)基因。结论在紫杉醇耐药及敏感细胞中,已知的耐药相关基因的表达差异很大,说明紫杉醇耐药机制是多因素、多通路相互复杂作用的过程,仍需要我们深入、全面、系统的研究、验证,才能最终转化为指导临床化疗方案的制定、有针对性的逆转耐药的方法。

The expression of resistance-related genes in ovarian cancer paclitaxel-resistancer cell after paclitaxel chemotherapy in vitro

Abstract:Objective To investigate expression of several drug resistance related genes between sensitive and resistant ovarian cancer cell lines after paditaxel chemotherapy in vitro.Methods RT-PCR method was used to detect the expressions of several drug resistance related genes between ovarian cancer cell A2780 and paelitaxel-resistant A2780/TS.Results In paclitaxel-resistant cell line A2780/TS,the inhibit rate was much lower than that in sensitive cell line A2780(P<0.01).The resistance index(RI)of A2780/TS cell was 3.650.There was no significant differencesinG1、G2、and S ratio between the A2780 and A2780/TS cells(P>0.05).Among those resistance related genes,multiple drug resistance(MDR-1)gene,P53 were expressed at lower leveh in resistant A2780/TS cells than in sensitive A2780 cells;while cytochrome P450 1B1(CYP1B1)and apoptotic inhibit protein survivin were expressed higher,and no obvious alterations were found concerning glutathione S transforase-pi(GST-Pi)and multiple drug resistance related protein-1(MRP-1).Conclusion The expression of known drng resistance related genes were distantly different in paclitaxel resistant and sensitive cell lines,suggesting more than one pathway leading to resistance transformation.It is necessary to study and validate widely,deeply and completely for setting down clinical chemotherapy method and reversing arug resistance.
