摘    要:目的:通过观察刺老苞根皮黄酮对新西兰兔骨性关节炎模型的软骨细胞增殖与凋亡基因表达的影响,初步探索其干预软骨细胞代谢的作用机制。方法:取5月龄新西兰兔骨性关节炎模型的膝关节软骨建立软骨细胞体外培养体系,分别采用MrITll法、TUNEL法、免疫组化法对细胞增殖、凋亡及细胞周期调控基因Bax、Bcl-2、Caspase-3、p53mRNA的表达进行检测。结果:不同剂量组刺老苞根皮黄酮可显著促进软骨细胞体外增殖,并可通过减弱软骨细胞Bax、Caspase-3、p53mRNA的表达,增强Bcl-2mRNA的表达来减缓软骨细胞凋亡进程。结论:刺老苞根皮黄酮可有效调控兔骨性关节炎软骨细胞代谢,增强细胞活性,延缓凋亡,具有研发成为骨性关节炎治疗药物的潜能。

Abstract:Objective:In this paper,it was studied about the effect of chondrocytes proliferation and apoptosis on cartilage cells by Aralia echinocauIis Hand.Mazz Flavonoids(AHF),which might light some suggestions for the control mechanism of cartilage cells by AHF Methods:Fifteen New Zealand white rabbits of 5 month old were selected in this study.Animal model was established by method of osteoarthritis animal model.The anterior and posterior cruciate ligament and medial collateral ligament were cut off and medial meniscus were cut articular cartilage cell cultured in vitro.Cells in the sham operation group was the normal control group,osteoarthritis cartilage cells in the model groups were added AHF 10,20,30 μg/ml respectively.A group of animals were sacrificed every week form the ninth weeks and the cartel age cells were isolated and cultured.The analysis of chondrocytes proliferation was used by MTT,and the analysis of chondrocytes apoptosis was used by TUNEL.Moreover,the expressions of Bax、Bcl-2、Caspase-3、p53 mRNA were deterred by immunochemical methods.Results: The chondrocytes proliferation of cartilage cells was increased by culturing with AHF in different dosages.And the chondrocytes apoptosis of cartilage cells was decreased by controlling Bax、Bcl-2、Caspase-3、p53 mRNA expression.Conclusion: The AHF has a function of active effect of osteoarthritis cartilage cells and inhibit apoptosis within a certain dose range.It may be the main mechanism for Aralia echinocauIis Hand.Mazz flavonoids to treat osteoarthritis.
