131I-Herceptin对乳腺癌细胞株的体外杀伤效应

目的 研究核素¹³¹I标记Herceptin对Her-2表达阳性乳腺癌细胞株的特异性杀伤作用。为放射免疫导向治疗奠定基础。方法 应用IODO-GEN方法将¹³¹I标记于抗Her-2单克隆抗体Herceptin,用MTT法检测¹³¹I-Herceptin对SK-BR-3、MCF-7、A5493种细胞株体外生长的抑制作用。结果 ¹³¹I-Herceptin的免疫活性与Herceptin无差异;¹³¹I-Herceptin对Her-2表达阳性的肿瘤细胞株的杀伤作用较Herceptin、¹³¹I明显增强(P<0.05),对于Her-2表达阴性或弱阳性的肿瘤细胞株无明显杀伤作用。结论 ¹³¹I-Herceptin可特异性杀伤Her-2表达阳性的肿瘤细胞。

Killing effect of 131I-Herceptin on breast cancer cell lines in vitro

Objective To study the specific killing effect of 131I-labeled Herceptin on breast cancer cell lines positive for Her-2 expression. Methods Anti-Her-2 monoclonal antibody Herceptin was labeled with 131I by means of IODO-GEN, and the killing effects of Herceptin on SK-BR-3, MCF-7 and A549 cell lines expressing Her-2 were evaluated with MTT assay. Results 131I-Herceptin had immunoactivity had to that of Herceptin but possessed stronger killing effects on the cell lines expressing Her-2 than Herceptin and 131I used alone (P<0.05). Its killing effect on cell lines negative or only weakly positive for Her-2 expression was similar to that of 131I and Herceptin. Conclusion 131I-Herceptin can specifically kill Her-2-positive cancer cells in vitro.
