原人参三醇组皂苷和人参皂苷-Re对MPP+诱导的SH-SY5Y细胞损伤的保护作用

摘    要:目的:探讨原人参三醇组皂苷(protopanaxatriol group,PPT)和人参皂苷-Re对1-甲基-4-苯基-吡啶阳离子(1-methyl-4-phenylpyridiniumion,MPP+)诱导的人多巴胺能神经母细胞瘤系SH-SY5Y细胞损伤的保护作用。方法:用不同浓度的PPT和人参皂苷-Re预处理细胞,以及不同体积分数的PPT和人参皂苷-Re含药血清预处理细胞,采用噻唑蓝(methylthiazolyldiphenyl-tetrazolium bromide,MTT)法检测细胞活力,计算细胞存活率,比较各组间的差异。结果:5μmol·L^-1和10μmol·L^-1的PPT和人参皂苷-Re均可以显著增加细胞存活率(与模型组比较,P〈0.05),体积分数10%的PPT和人参皂苷-Re含药血清能显著增加细胞存活率(与模型组比较,P〈0.001)。结论:PPT和人参皂苷-Re对MPP+诱导的SH-SY5Y细胞损伤有显著的保护作用,具有潜在的抗帕金森病作用。

Abstract:Objective:To investigate the protective effect of PPT (protopanaxatriol group) and ginsenoside-Re on l-methyl-4-phenylpyridiniumion (MPP+) -induced damage in SH- SY5Y (human dopaminergic neuroblastoma cell line) cells. Metbods:SH-SY5Y cells were treated with different concentrations of PPT,ginsenoside-Re or MPP containing serum. MTT colorimetry was used to test the viability of the cells and the survival rates were calculated and analyzed. Results:Cells treated by either 5 μmol·L^-1, 10 μmol·L^-1 PPT or ginsenoside-Re showed significantly higher viability as compared to control group (P〈0.05). In addition, 10% PPT treated or ginsenoside-Re medicated serum treated groups showed similar protective effect.Conelusion.PPT and ginsenoside-Re have neuroprotective effects in a cell model of Parkinson's disease MPP~ -induced SH-SYSY cell death, suggesting that both PPT and ginsenoside-Re may have anti-PD effect.
