山楂总黄酮对酒精诱导小鼠酒精性肝病的影响

摘    要:目的 研究山楂总黄酮对酒精诱导小鼠酒精性肝病的影响.方法 以56%体积分数白酒(1.5ml·100g-1)体重造模6周后,随机分组,山楂黄酮低、中、高剂量组(5,10,20g·kg1)和联苯双酯组(0.6g·kg1),在酒精继续灌胃的基础上,按剂量给药,1次·d-1,共12周.检测小鼠血清和肝组织中丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)、超氧化物歧化酶(SOD)活性及丙二醛(MDA)、一氧化氮(NO)、谷胱甘肽过氧化物酶(GSH-PX)、肝匀浆中肿瘤坏死因子(TNF-α)含量.结果 山楂总黄酮对酒精诱导小鼠酒精性肝病形成后SOD、GSH-PX的活性含量有回升作用,并能阻止与肝病相应的ALT、AST活性及降低MDA、NO、TNF-a含量.结论 山楂总黄酮对酒精诱导小鼠酒精性肝病有预防作用,可能与其抗氧化和免疫调节作用有关.

Abstract:Objective To study the effect of total flavonoids of hawthorn on ethanol induced alcoholic liver disease in mice. Methods Animal model was established by 56% (v/v) (1.5ml · 100^-1 ) body,which were divided hawthorn flavonoids of low, middle and high dose group,(5, 10, 20g · kg^-1 ) and bifendate group (0. 6g · kg ^-1 ) after 6 weeks. Every group was respectively treated for 12 weeks with total flavonoids of hawthorn, bifendate or normal saline accordingly. The activities of alanine transaminase (ALT) aspartate transaminase (AST) in serum , superoxide dismutase ( SOD) ,malondialdehyde (MDA) ,nitric oxide (NO) ,glutathione peroxidase (GSI-t-PX) in liver homogenate, tumor necrosis factor (TNF-a) content were investigated. Results otal flavone of hawthorn can increase activity of SOD,GSH-PX,diseaseing ALT,AST activity and MDA,NO,TNF a content. Conclusion Hawthorn flavonoids have a preventive effect on alcoholic liver disease in mice induced by alcohol, which may be related to its antioxidant and immune regulation
