子宫内膜异位症患者腹水与血清中肝细胞生长因子的检测

摘    要:扩散因子(肝细胞生长因子)(SF/HGF)是一种多功能的生长因子,与肿瘤细胞的增生、血管生成、侵袭性密切相关。本研究的目的是探索HGF在子宫内膜异位症发病机制中的作用及HGF与子宫内膜异位症发生发展的关联。本研究应用酶联免疫吸附反应技术检测了126位妇女(其中72位为子宫内膜异位症患者,54为正常对照)血清及腹水中HGF浓度。与对照组比较,Ⅰ/Ⅱ和Ⅲ/Ⅳ子宫内膜异位症患者血清和腹水中的HGF浓度升高,3组妇女腹水中HGF浓度分别为(0.337±0.065)、(0.458±0.160)、(0.915±0.113)ng/ml 。血清中HGF浓度分别为(0.335±0.663)、(0.456±0.151)、(0.907±0.17)ng/ml。Ⅲ/Ⅳ子宫内膜异位症患者血清和腹水中HGF浓度与Ⅰ/Ⅱ期患者和对照组比较,差异有统计学意义。三组妇女血清与腹水的HGF浓度均呈正相关。HGF浓度与月经周期无明显相关。处于同一月经周期的患者与正常妇女血清及腹水HGF浓度比较,显示HGF浓度的升高主要是子宫内膜异位症的发生和发展相关,与月经周期无明显相关。

Abstract:Scatter factor (hepatocyte growth factor, SF/HGF) is a multifunctional polypeptide growth factor that plays animportant role in tumor proliferation, angiogenesis and invasiveness. The aim of this study was to investigate the role of HGFin endometriosis and their relationship. Peritoneal fluid and serum samples of 126 women of reproductive age receivinglaparoscopy (72 with endometriosis and 54 without endometriosis) were obtained and enzyme-linked immunosorbent assay(ELISA) was employed to measure HGF concentrations. The results revealed that the concentrations of HGF were significantlyelevated in women with endometriosis stages Ⅰ/Ⅱ and Ⅲ/Ⅳas compared with HGF level in the control group (peritoneal fluidlevels of 0.337±0.065, 0.458±0.160, 0.915±0.113, P<0.001; serum level of 0.335±0.663, 0.456±0.151, 0.907±0.170, P<0.001respectively). A positive correlation was found between peritoneal fluid and serum levels of HGF, which varied little withphases of the menstrual cycle. Comparisons of the women in the same menstrual cycle between the three groups revealed thatincreased levels of HGF in patients with advanced endometriosis were not attributed to the cycle phase. Our results indicatedthat the degree of endometriosis was positively correlated with HGF concentration in peritoneal fluid and serum. Therefore,the detection of serum HGF concentration may be useful in classifying endometriosis, and may also help in selecting theoptimal treatment methods.
