摘    要:目的:制备rhBMP-2-PLA纳米微球缓释系统,观察其对兔成骨细胞的生物学效应。方法:采用复乳-干燥法制备rhBMP-2纳米微球,观察其一般特性,并模拟体内条件研究BMP-PLA纳米微球的体外缓释特性。采用MTT法检测微球对成骨细胞增殖状况的影响,并与单纯rhBMP-2的作用进行比较。结果:纳米微球表面光滑圆整,球体大小均匀,平均粒径为45 nm,rhBMP-2-PLA纳米微球的包封率和载药量分别为(90.67±1.23)%和(134.65±0.44)×10-3]%,微球体外释药符合Higuichi方程。该微球缓释系统有生物活性,能显著促进兔成骨细胞的增殖,其效应高于单纯施加rhBMP-2的效应。结论:rhBMP-2-PLA纳米微球缓释系统的作用明显优于单纯rhBMP-2,在骨创伤的治疗中有良好的前景。

Abstract:Objective: To fabricate recombinant human bone morphogenetic-2 polylactic acid(rhBMP-2-PLA) sustained release nanospheres,and evaluate its biological effects on cultured rabbit osteoblasts.Methods: The rhBMP-2-PLA nanospheres were prepared by w/o/w multiple emulsion volatilizing method.General properties were observed.The rhBMP-2-PLA nanospheres release characteristics were studied in simulated test.Cell culture technique and MTT colorimetric assay were used to evaluate the proliferation of the cells cultured with the rhBMP-2-PLA nanospheres or rhBMP-2.Results: The rhBMP-2-PLA nanospheres were round particles with smooth surface.The uniform spheres with a mean particle size of 45nm.The drug loading amount and encapsulation efficiency of rhBMP-2-PLA nanospheres were(90.67±1.23)% and (134.65±0.44)×10-3]% respectively.The release profile of rhBMP-2-PLA nanospheres in vitro was figured by Higuichi equation.Statistical analysis showed that rhBMP-2-PLA nanospheres in vitro exhibited biological effects,which could significantly enhance proliferation of rabbit osteoblasts.Biological effect of rhBMP-2-PLA nanospheres on osteoblasts was significantly higher than that of rhBMP-2.Conclusion: The rhBMP-2-PLA nanospheres have excellent biological effects compared with common rhBMP-2,and appear to have a good prospect in clinical orthopedics.
