摘    要:用AOH体外诱导不同时间(24小时,1周和3周)的人胎儿食管上皮组织,提取该组织高分子量的DNA,从10例食管癌组织及癌旁“正常”组织中分别提以DNA,用EcoRI酶切,Southern转移,与EGFr基因探针杂交,结果显示:AOH诱导24小时人胎儿食管上皮中即有EGFr基因扩增,并一直持续到第三周,2例食管癌组织,1例癌旁“正常”组织也有EGFr基因扩增,因此:AOH可以激活EGFr。EGFr基

Abstract:High-molecular-weight DNA from the human fetal esophagusl (HFE) induced by alternariol(AOH) in vitro at different times(24 hours, 1 week and 3 weeks)in vitro was purified. Meanwhile it was isolated from 10 tumours and non-tumour adjacent tissues of the esophagus obtained from the patients who have undergone surgery for esophageal cancer. These DNA samples were also digested with EcoRI enzyme and subjected to southern blotting. The southetn blots was probed with EGFr gene. The result showed that EGFr gene in HFE induced by AOH in 24 hours has amplified and it couti- nued to the third week.The amplification of EGFr also existed in the two cases of esopha- geal cancer tissues and one case of non-tumour adjacent tissues of the esophagus, There- fore, EGFr can be activated by AOH. The activation of EGFr gene is probably due to the initiating change of the esophageal cancer and it played an important part in the genesis and deyelopment of the esophageal cancer.The amplification of EGFr in the non- tumour adjacent tissue should go beyond the present range of pathological diagnosis
