甲状旁腺素促成骨样细胞增殖作用及其与钾离子通道的关系

摘    要:为研究甲状旁腺素 (PTH)促大鼠成骨样肉瘤细胞 (UMR10 6 )增殖作用及与钾离子通道活动的关系。采用磺基罗丹明(SRB)染色法观察细胞增殖改变并用膜片钳技术记录细胞膜钾离子通道电流的变化。同时用 3H- Td R掺入法测定 DNA合成的改变。结果显示 ,PTH可以明显地促进 UMR10 6细胞的增殖 ,且使 3H- Td R掺入率增加 77.14 %。在含有 PTH(10 - 8mol· L- 1 )的培养液中培养 12 h后 ,钾通道电流比对照组有明显增加 ;而且用钾通道阻断剂后 ,3H- Td R掺入率下降。表明其可促进大鼠成骨样肉瘤的 DNA合成和细胞增殖 ,且细胞膜钾通道的活动与此促增殖活动密切相关

Abstract:Objective: To study the effect of on PTH on cell proliferation in rat osteoblast like cells line and the relationship of K channel and the proliferation. Methods: The cells were counted with SRB staining technique. And the way of the Patch clamp was used for recording the activity of K channel. At the meantime the DNA synthesis was identified with the method of 3H TdR incorporation. Results: Call proliferation could be demonstrated when the cells were exposed to PTH. Cell count increased by 50.00% increase 3H TdR incorporation by 77.14% at the physiological concentrations. PTH was added to culture medium for 12 hours, these K currents increase remarkably than the control. Addition of K channel blocker TEA at the various times after addition of PTH, the 3H TdR incorporation could be blocked to certain degree. Conclusions: PTH was the patent effector on the UMR106 cells proliferation and could increase DNA synthesis of the cells, This process may be involved in the activity of the K channel.
