转基因牛乳中重组人乳铁蛋白的仿生亲和纯化

摘    要:目的从转基因牛乳中亲和纯化重组人乳铁蛋白。方法从仿生亲和介质库中筛选亲和配体,进行重组人乳铁蛋白的纯化研究。结果筛选出可以特异性结合重组人乳铁蛋白的固定化β-丙氨酸亲和配体,从转基因牛乳中一步纯化的重组人乳铁蛋白纯度为95.2%,蛋白回收率可达81.9%。经放大实验表明该亲和纯化工艺适合工业上大规模纯化乳铁蛋白。结论固定化β-丙氨酸可从转基因牛乳中纯化出高纯度重组人乳铁蛋白,工艺适合工业化生产。

Abstract:Purpose To purify the recombinant human lactoferrin from the milk of transgenic cows by affinity chromatography.Methods A library of biomimetic affinity ligands was screened for lactoferrin binding specificity and capacity.Affinity chromatography was carried out based on the synthetic ligand selected.Results Immobilised β-Alanine was found to be an affinity ligand specific for recombinant human lactoferrin.By this ligand,recombinant human lactoferrin was one-step purified from the milk of transgenic cows with a purity of 95.2% and the protein recovery could reach 81.9%.Conclusion Immobilised β-Alanine can be used to purify recombinant human lactoferrin,and this process was scalable and suitable for commercialization.
