血管紧张素Ⅱ受体拮抗剂对阿霉素肾病模型骨调素表达的影响

摘    要:目的 探讨骨调素 ( OPN)在阿霉素肾病模型中的表达和作用以及血管紧张素 ( Ang )受体拮抗剂对其表达的影响。方法 将实验动物随机分为正常对照组 ( C组 ) ,阿霉素肾病组 ( A组 )和厄贝沙坦治疗组 ( IR组 ) ,第2、 4周用免疫组化方法检测各组肾内 OPN和单核巨噬细胞抗原 ( ED1)的表达情况。结果 发现在第 2周和第 4周 ,A组和 IR组 OPN与 ED1表达水平均明显高于正常组 ( P<0 .0 5 ) ,而 IR组 OPN与 ED1表达水平均低于 A组 ( P<0 .0 5 )。在第 2周和第 4周 ,A组 OPN与 ED1的表达水平呈显著正相关 ( r=0 .92 2 ,P<0 .0 1;r=0 .80 0 ,P<0 .0 5 )。结论  OPN在阿霉素肾病这种微小病变模型中是一种重要的趋化因子 ,与肾小管间质单核巨噬细胞浸润密切相关 ,使用 Ang 受体拮抗剂可使该模型 OPN表达下调 ,从而减轻了炎性细胞浸润和肾损伤

Abstract:Objective To investigate the expression and potential role of osteopontin (OPN) in rat adriamycin-induced nephropathy (AN) model and the effects of Irbesartan on them. Methods The rats were divided into the following groups: normal control (C), AN (A) and Irbesartan-treated (50 mg/kg every day, IR group). Immunohistochemical method was used to detect the expression of OPN and ED1 at 2nd and 4th week. Results Compared to the group C, there was a significant increase in expression of OPN and ED1 in cortical tubular epithelium during the development of AN in group IR and group A (P<0.05). The expression levels of OPN and ED1 in group IR were significantly lower than in group A (P<0.05). On the week 2 and 4, the expression of OPN and ED1 was closely correlated (r=0.922, P<0.01; r=0.800, P<0.05). Conclusion OPN is an important chemokine in AN rats with minimal change, and has correlation with macrophage infiltration in tubulointerstitial areas. Irbesartan has some renal protective effect on AN rats, partly through down-regulating OPN expression and reducing the macrophage infiltration.
