摘    要:目的了解人红细胞6-磷酸葡萄糖脱氢酶(G6PD)活性与红细胞还原型谷胱甘肽含量的关系及对红细胞脆性的影响。方法用改良G6PD定量比值法测定G6PD活性,用Beutler改良法测定红细胞GSH,采用Habermann&Hardt法测定溶血率;用SPSS13.0进行t检验及相关性分析。结果在G6PD活性正常的红细胞中,GSH含量与G6PD活性呈正相关,两者之间的相关系数为0.7154,R2=0.5118;G6PD缺乏的红细胞内的GSH含量偏低;与G6PD活性正常红细胞比较,G6PD缺乏红细胞对H2O2氧化损伤更敏感(P<0.01)。结论 G6PD缺乏患者的血液中GSH含量偏低,影响其抗氧化能力。

Abstract:Objective To study the relationship between the glucose-6-phosphate dehydrogenase (G6PD) activity and the glutathione (GSH) levels in human erythrocytes, and investigate the effect of G6PD activity on the fragility of erythrocytes. Methods G6PD activity was measured by modified quantitative G6PD/6-GP ratio method. The GSH levels in erythrocytes were measured by modified Beutler methhod. The hemolytic rates of erythrocytes were measured by method of Habermann and Hardt. The correlation analysis and t-test were done by using SPSS13.0. Results The GSH levels showed high correlation to G6PD activity, with a correlation coefficient of 0.715 4 and R2 of 0.511 8. The erythrocytes with G6PD deficiency showed significantly reduced GSH levels than the erythrocytes with normal G6PD activity. Besides, erythrocytes with G6PD deficiency are more vulnerable to the oxidant stress of H2O2. Conclusion Erythrocytes with G6PD deficiency have lower GSH levels and antioxidant capacity.
