人类表皮生长因子受体2血管内皮生长因子在胃癌组织芯片中的表达及意义

摘    要:目的探讨人类表皮生长因子受体2(HER2)和血管内皮生长因子(VEGF)蛋白在388份胃癌组织芯片中的表达,分析其与患者临床病理学参数之间的关系及意义。方法在胃癌组织芯片上,应用免疫组织化学SP法检测HER2和VEGF蛋白表达水平,分析HER2和VEGF在胃癌组织的表达与患者临床病理特征的关系。结果 HER2在胃癌组织中的阳性表达率为9.3%(36/388);VEGF的阳性表达率为28.9%(112/388)。HER2的表达在肿瘤不同分化程度、Lauren分型间差异有统计学意义(P<0.05);VEGF的表达在肿瘤不同TNM分期、淋巴结转移间差异有统计学意义(P<0.05),且HER2和VEGF之间呈负相关(r=-0.140,P=0.022)。结论同时检测胃癌组织中的HER2和VEGF蛋白的表达对判断肿瘤的恶性程度和估计预后可能有一定意义。

Abstract:Objective To evaluate the expression and significance of HER2 and VEGF protein in human gastric carcinoma tissues in 388 cases,and to study its clinical significance and the association with the clinicopathological features in tissues-microarray.Methods The expression levels of HER2 and VEGF protein were measured in human gastric carcinoma tissues by SP immunohistochemical method in tissue-microarray.The relationship between their expression and clinicopathological features was studied.Results Positive expression of HER2 protein was found in 9.3%(36/388)cases of gastric carcinoma,the positive rate of VEGF expression was 28.9%(112/388),respectively.The expression of HER2 was significantly correlated in gastric carcinoma to Lauren’s classification and differentiation.The overexpression of VEGF had a significant relationship with lymph node metastases and TNM stages.HER2 had intimate negative relationship with VEGF(r=-0.140,P=0.022).Conclusion Effective detection of HER2 and VEGF proteins may contribute to the evaluation of malignancy and biological behavior of gastric cancer.
