大鼠局灶性脑缺血模型制作方法的改进

摘    要:目的 探索导致栓线法局灶性脑缺血模型制作失败的原因,提出相应的技术解决方法。方法 按照文献方法,采用体重为250~350g的SD大鼠进行栓线法局灶性脑缺血模型制作。结果 在模型制作过程中,遇到的最大困难是栓线插线时的大出血,以及有时栓线无法正确插入颈内动脉。不用血管夹夹闭颈内、外动脉,直接在颈内动脉靠近分叉处套上一根丝线,用血管钳牵拉,就可以很容易地控制出血;遇到栓线插入困难时,向深层次解剖,显露翼腭动脉和颈内动脉分叉,就可以直视下将栓线插入颈内动脉。结论 采用预先在颈内动脉套线,可以方便有效地控制出血。插线困难时,向深部解剖显露翼腭动脉后可以在直视下将栓线送入颈内动脉,无需结扎翼腭动脉。整个手术操作可以由一个人完成。

Abstract:Objective To reduce the bleeding and shorten the length of the procedure of producing focal cerebral ischemic models by suture method.Methods Focal cerebral ischemia models were made inSprague-Dawley rats weighing 250-350 g in accordance with Koizumi's and Longa's methods. Results Massive bleeding and some times the threads couldn't be inserted into the internal carotid artery (ICA) were the two major difficulties for making ischemic models points. To reduce the rebleeding , we tie the ICA loosely with a 6-0 silk suture which was retracted by a vascular forceps. The pterygopalatine artery is dissected and divided only when the thread is difficult to be inserted into ICA Once the origin of the pterygopalatine artery was dissected the thread can be easily inserted into ICA. Conclusion A 6-0 silk suture was tied loosely around the ICA , through which the bleeding can be controlled wonderfully. The pterygopalatine artery shoud be dissected only when the thread can not be inserted into ICA correctly and the ligation of this artery is not necessary at anytime.
