大鼠脑组织中氨基酸类神经递质的rP-HPLC-UV柱前衍生定量分析

摘    要:目的:测定大鼠脑组织中纹状体、海马、皮质中氨基酸的含量,研究中枢神经系统及脑能量代谢和某些疾病之间的关系。方法:用反相高效液相色谱法测定脑组织中的8种游离氨基酸,以2,4-二硝基氟苯(DNFB)作为衍生剂,在UV360nm进行测定,以外标法进行计算。结果:在20min内分出8种氨基酸,分离完全,且纹状体氨基酸含量高于海马和皮质。结论:本方法简便,灵敏度高,分析时间短,适用于一般实验室。

Abstract:Objective:To determine the content of amino acid in corpus striatum, hippocampus and cortex of rat brain tissue, investigate the relationship of central nervous system, brain energy metabolism and certain diseases Methods: We analyzed 8 free amino acids by reversed phase high performance liquid chromatography The samples were derivated with DNFB ,detected with an UV detector at 360nm and calculated by outer criteria Results: This method can determine 8 amino acids in 20 minutes, and completely separate them We found that the content of amino acid in corpus striatum is higher than that of in hippocampus and cortex Conclusions: This method is simple, rapid and high sensible We believe it can be used successfully in common laboratory
