川芎嗪注射液抑制子宫内膜异位症微血管密度的实验研究

摘    要:目的:观察川芎嗪注射液(TMP)对大鼠子宫内膜异位症微血管密度的影响和抗血管形成作用。方法:建立Wistar大鼠子宫内膜异位症动物模型,4周后将建模成功的大鼠随机分为对照组、ip TMP 50,100,200 mg/(kg.d)连续给药15d,SP免疫组化法测定在位内膜和异位病灶组织中MVD的表达。结果:TMP治疗EMs模型大鼠4周后异位组织中MVD的表达明显低于对照组,差异有显著意义(P〈0.05),而在位内膜中MVD的表达与对照组相比,差异无显著意义(P〉0.05);TMP可剂量依赖性的抑制EMs异位组织的增生(P〈0.05)。结论:TMP对子宫内膜异位症大鼠异位组织中MVD的表达和新生血管形成均有抑制作用,作用呈剂量依赖性。TMP有望成为子宫内膜异位症抗血管形成治疗的新药。

Abstract:Objective:To observe the effect of TMP on micro-vessel density(MVD) and antiangiogenesis in rat endometriosis.Methods:Wistar rat endometriosis models were established,after 4 weeks,the successful endometriosis rats were divided into control group,TMP50mg·kg-1·d-1group,TMP100mg·kg-1·d-1group and TMP200 mg·kg-1·d-1group and the rats were given successive intragastric administration for 15 days.Ectopic endometrium graft volume was observed and MVD was determined by immunostaining for CD34.Results:MVD in endometriosis was decreased remarkably after 4-week treatment with TMP(P0.05),with was no significant difference.TMP could significantly inhibit proliferation of ectopic endometrium in a dose-dependent manner.Conclusion:TMP has an inhibitory effect on the MVD expression and angiogenesis in rat endometriosis in a dose-dependent manner.TMP may become a new drug of antiangiogenesis in endometriosis.
