胸腺五肽脂质体的处方及制备工艺研究

摘    要:目的研究胸腺五肽脂质体的处方筛选及制备工艺。方法采用逆向蒸发法制备胸腺五肽脂质体,超速离心法测其包封率,以包封率和粒径为指标,采用正交试验设计法优化处方,并对其表面特征、包封率、粒径等情况进行考察。结果最佳处方制备的胸腺五肽脂质体平均粒径为108.2 nm,药物的平均包封率为62.10%,Zeta电位为-48 mV,脂质体外观圆整均匀,分散性好。结论所制胸腺五肽脂质体工艺简单、重现性好,粒径小,包封率较高,为其进一步研究奠定了基础。

Abstract:Purpose To study the optimal formulation and preparation of thymopentin liposome.Methods Reverse-phase evaporation technique was chosen for the preparation of thymopentin liposome and entrapment efficiency was measured by ultracentrifugation.Orthogonal design was used to optimize the formulation by taking encapsulation efficiency and the particle size as indexes.The properties of liposome including morphology,entrapment efficiency,mean diameter,etc were studied.Results Mean diameter and encapsulation efficiency of the prepared thymopentin liposome were 108.2 nm and 62.10%,respectively.The Zeta-potential was-48 mV.The liposome was smooth and spherieal in appearance with better dispersion.Conclusion Preparation technique is simple and reproducible.The thymopentin liposome prepared in this study has smaller size and higher encapsulation efficiency.It was worthy to be further studied.
