姜黄素对肝癌HepG2和Bel-7404细胞增殖的抑制作用

摘    要:目的研究姜黄素对人肝癌HepG2和Bel-7404细胞增殖的影响。方法不同浓度的姜黄素(2.5,10.0,25.0,50.0,100.0μg/mL),0.2%二甲基亚砜的RPMI 1640培养基加细胞为空白对照,2μg/mL顺铂为阳性对照。MTT法检测肝癌HepG2和Bel-7404细胞的增殖。结果姜黄素对肝癌细胞HepG2和Bel-7404的增殖抑制有量效关系,并呈时间依赖性。在相同作用时间下,姜黄素组、顺铂组对肝癌细胞HepG2和Bel-7404增殖有明显的抑制作用,与空白对照组比较均有显著性差异(P<0.05)。与空白对照组比较,2.5,10.0μg/mL姜黄素对肝癌细胞HepG2和Bel-7404的增殖没有明显的抑制作用(P>0.05),而25.0,50.0,100.0μg/mL姜黄素以及顺铂组对肝癌细胞HepG2和Bel-7404的增殖均有明显的抑制作用(P<0.05)。结论姜黄素可以明显的抑制肝癌细胞HepG2和Bel-7404的生长,且存在剂量-时间的关系。

Abstract:Purpose To investigate the anti-proliferative effects of curcumin on human hepatocellular carcinoma HepG2 and Bel-7404 cells.Methods In this experiment were set up the groups of different concentrations curcumin(2.5,10.0,25.0,50.0 and 100.0 μg/mL),control group(0.2% DMSO+RPMI 1640 medium+ cells) and 2 μg/mL cisplatin group.The hepatocellular carcinoma HepG2 and Bel-7404 cells were detected by MTT method.Results The anti-proliferation of curcumin had a dose-effect relationship and was in time-dependent manner.Under the condition of the same reaction time,the anti-proliferative effects of curcumin and cisplatin were different significantly(P<0.05).Compared with control group,the anti-proliferative effects of curcumin in 2.5 and 10.0 μg/mL had no obvious effects(P>0.05),and its anti-proliferative effects in 25.0,50.0,and 100.0 μg/mL were significantly different(P<0.05).Conclusion Curcumin could significantly inhibit the proliferation of human hepatocellular carcinoma HepG2 and Bel-7404 cells in vitro.
