吴茱萸次碱对高血压大鼠血压的影响及作用机制

摘    要:目的观察吴茱萸次碱对高血压模型大鼠血压的影响,并初步探讨其作用机制。方法分别采用自发性高血压大鼠(SHR)及两肾一夹(2K1C)高血压大鼠模型,设正常对照组、模型组、卡托普利阳性组及吴茱萸次碱低、中、高剂量组(10,20,40 mg/kg),每日灌胃给药1次,给药4周,每周测定1次血压,末次给药后,腹主动脉取血,分别测定血浆血栓素(TXB2)、6-酮-前列腺-F1α(6-Keto-PGF1α)、肾素活性(PRA)、心房钠肽(ANP)水平。结果在两个模型中吴茱萸次碱均有较好的降压作用,并能使SHR大鼠血浆TXB2水平降低,6-Keto-PGF1α水平升高;使2K1C大鼠血浆PRA、ANP水平升高。结论吴茱萸次碱能明显降低SHR及2K1C大鼠血压,其降压效果可能通过调节PGI2和TXA2水平,改善血管内皮功能及增加舒血管物质ANP水平实现的。

Abstract:Purpose To investigate the antihypertensive effect and mechanism of rutaecarpine(Rut) in hypertensive rats.Methods The spontaneously hypertensive rats(SHR) and two-kidney,one-clip(2K1C) hypertensive rats were used in our research.After dividing into normal group,model group,positive group and Rut(10,20,40 mg/kg),Rats of the drug groups were intragastrically administrated the corresponding drugs by once daily for 4 weeks,and pressure was measured once a week.After the last administration,the blood was collected from abdominal aorta to measure the plasma thromboxane B2(TXB2),6-Keto-PGF1α,plasma renin activity(PRA),and atrial natriuretic peptide(ANP).Results Rut can decrease blood pressure in both SHR and 2K1C hypertensive rats,it can also reduce the TXB2 level,increase the 6-Keto-PGF1α level in SHR rats,and increase the PRA and ANP level in 2K1C rats.Conclusion The Rut can effectively decrease blood pressure.It′s antihypertensive effect may be related to the regulation of plasma PGI2 and TXA2 level and the increase of plasma ANP.
