旱黑口服液对D-半乳糖致衰老小鼠运动能力的影响

摘    要:目的:探讨旱黑口服液对D-半乳糖致衰老小鼠运动能力的影响及机制.方法:昆明种小白鼠50只随机分为空白对照组、造模组、旱黑口服液(高剂量组、中剂量组、低剂量组)组5组,每组10只.除空白对照组外,其余各组均给予50g/L D-半乳糖0.3 ml颈背部皮下注射,连续给药42 d造模,同时旱黑口服液高、中、低剂量组分别给予旱黑口服液(20 g·kg-1·d-1,12 g·kg-1·d-1,8 g·kg-1·d-1)灌胃;其他组给予生理盐水(0.5 ml/d)灌胃,6周后测定小鼠游泳力竭时间、记忆能力、心肌、脑组织、臀大肌红肌组织SOD、Na -K ATP、Ca2 -Mg2 ATP、Ca2 、MDA水平.结果:旱黑口服液高、中剂量组游泳力竭时间长于造模组(P<0.01或0.05),记忆能力结果显示潜伏期延长(P<0.01),错误次数减少(P<0.01或0.05).旱黑口服液组小鼠心、脑组织中SOD、Na -K ATP、Ca2 -Mg2 -ATP活性较造模组升高(P<0.01或0.05),Ca2 、MDA含量降低(P<0.01或0.05).结论:旱黑口服液能防止D-半乳糖致衰老小鼠的运动能力下降、学习记忆障碍等衰老体征,其作用可能与其提高抗氧化酶活性、清除自由基、调节细胞内外Ca2 平衡有关.

Abstract:Aim: To observe the influence of Hanhei oral solution on the movement ability of the old and feeble mice caused by D-galactose.Methods: A total of 50 Kunming mice were divided into five groups: control group, model group, and Hanhei oral solution groups (high concentration group, middle concentration group and low concentration group). Except for the control group, the other 4 groups were given 50 g/L D-galactose for 42 d. Meanwhile, the Hanhei oral solution groups were given Hanhei oral solution at three different levels, respectively, and the other groups were given physiological saline.The exhausting time for swimming, memorial ability, SOD, Na -K -ATP,Ca 2 -Mg 2 -ATP, Ca 2 ,MDA in different tissue after 6 weeks were assessed. Results: The exhausting time for swimming in the high and middle concentration group was prolonged compared with that of the model group(P<0.01).The memorial ability test showed longer latency period. The levels of SOD, Na -K -ATP,Ca 2 -Mg 2 -ATP improved (P<0.01 or 0.05) and the levels of Ca 2 and MDA reduced(P<0.01 or 0.05) in Hanhei oral solution groups. Conclusion:Hanhei oral solution can effectively prevent the decrease of movement ability and memorial ability caused by D-galactose. The consequence might be related to the improvement of the activity of anti-oxidized enzyme, the elimination of the freebase, and regulation of the Ca 2 balance in and out the cells.
