摘    要:目的 考察Gadd 45基因在健康人群个体间表达的差异,评估Gadd 45发展成为一种新的辐射生物剂量计的可行性。方法 采集20例健康献血员血液样本,提取白细胞RNA进行反转录,以β-actin作为内对照基因,建立实时定量聚合酶链式反应(PCR)方法,并以此方法探讨人群中Gadd 45基因表达水平的差异。结果 20例受试者血液标本中的Gadd 45表达水平之间存在较大差异,且其表达水平与性别无关。结论 放射诱导基因Gadd 45的表达可能不适合做生物剂量计。

Abstract:Objective To investigate the difference of Gadd 45 expression in healthy population, and to evaluate the possibility of developing Gadd 45 expression into a new radiation bio-dosimeter. Methods Twenty blood samples from healthy population were collected and the total RNA was extracted from white blood cells and reverse transcribed into cDNA. The method of real-time PCR was used to detect the different level of Gadd 45 expression in human population, with β-actin as an internal control. Results There was a significant difference in Gadd 45 expression among 20 subjects. In addition, the difference was irrelevant to the sex. Conclusion The expression of irradiation-inducible gene Gadd 45 might not be a good bio-dosimeter.
