Abstract:Objective To investigate whether traffic related PM2.5 and its water-soluble components can induce spleen autophagy, activate NLRP3 inflammasome and eventually cause spleen injury in asthmatic rats. Methods In total 25 male Sprague Dawley rats were randomly divided into five groups, including normal saline group(NS group), asthma group (OVA group), PM2.5 suspension group (PM2.5 group), the group with low PM2.5 water soluble components (WSL group), and the group with high PM2.5 water soluble components(WSH group). The asthma model of rats was established by intraperitoneal injection of OVA and aluminum hydroxide suspension, and then stimulated by OVA atomization inhalation. The rats were killed after exposure to PM2.5 and its water-soluble components for 8 times, and then the spleen tissue was taken out for standby. The protein expression levels of LC3, P62, NLRP3, pro-caspase-1, caspase-1 p10 and IL-1β in spleen were determined by Western blot. The gene expression levels of ATG5, Beclin-1, NLRP3 and ASC in spleen were determined by qRT-PCR. Results The protein expression of NLRP3, pro-caspase-1, caspase-1 p10, IL-1β and ASC gene expression in WSH group and PM2.5 group were higher than those in NS group. Autophagy related genes ATG5 and Beclin-1 in WSH group were significantly higher than those in NS group and OVA group. P62 protein expression in NS group was significantly higher than that in other groups. Conclusion PM2.5 and its water-soluble components can induce autophagy and activate NLRP3 inflammasome in the spleen of asthmatic rats.
