Abstract:Objective To develop a chromogenic medium for Yersinia pestis based on the resistance of Yersinia pestis to certain inhibitors and the color development of enzymatic substrates, and to evaluate its detection effect. Methods Four Yersinia pestis color development media were prepared based on Yersinia pestis selective medium (CIN agar) with nutrients to promote the growth of Yersinia pestis and different concentrations of bacteriostatic agents, and the best Yersinia pestis color development media were screened by single-factor experiments. Results The 1% bile salt chromogenic medium screened by single factor had strong inhibitory effect on common pathogenic bacteria and good selectivity and specificity for Yersinia. Yersinia showed rosy red colonies after incubation at 28℃ for 18-24 h, and the colonies were typical and easy to be separated. Conclusion The optimized chromogenic medium of Yersinia pestis has good chromogenic effect, strong specificity and high sensitivity, which can improve the isolation rate of Yersinia pestis and is suitable for the primary screening of Yersinia pestis isolation culture.
