SUR1基因多态性与2型糖尿病的相关性研究

目的：探讨2型糖尿病（T2DM）与磺脲类药物受体1（SUR1）基因多态性的关系。方法：随机抽取正常人136例（正常组）及2型DM患者173例（T2DM组）外周血白细胞基因组DNA，PCR扩增SUR1基因31号外显子多态性位点的区域．用限制性酶切片段长度多态性分析其基因型，并用测序证实不同酶切图形基因型的准确性。结果：T2DM组和正常组SUR1基因31号外显子基因型及等位基因分布差别有统计学意义（P〈0．05）。结论：SUR1基因可能是中国天津地区2型糖尿病发生的易感基因。

Correlation between Polymorphism of Sulfonylurea Receptor 1 Gene and Type 2 Diabetes

Objective: To study whether the polymorphism of human sulfonylurea receptor (SUR1) gene correlated with type 2 DM in Tianjin. Methods: One hundred and thirty six normal subjects and 173 patients with type 2 DM in Tianjin were included. Genetic DNA was extracted from white cells of peripheral blood. The aim fragment containing polymorphism site was amplified by PCR and the gene type was analyzed by the lenght of restriction enzymes. Furthermore the accuracy of genotypes was analyzed through the sequence. Results: The position of SUR1 gene exou31 and allele were found to show significant difference between type 2 DM patients and normal subjects. The mutation frequency in exon 31 in normal subjects was significantly higher than that in patients with type 2 DM (P<0.05). Conclusion: The polymorphism site of exon 31 within human SUR1 gene is maybe correlated with population with type 2 DM in Tianjin. SUR1 is a possibly susceptible gene for type 2 DM in Tianjin.
