谷氨酸诱导骨髓间充质干细胞毒性损伤的作用

摘    要:目的研究谷氨酸对骨髓间充质干细胞(BMSCs)毒性损伤的作用。方法原代培养大鼠BMSCs,流式细胞仪分析细胞表型,四甲基氮唑蓝(Mrrr)法检测分别经10、30、50mmol/L的谷氨酸作用24h后的BMSCs存活率,并用PI/Annexin—V双染法流式细胞仪计算各组凋亡细胞的比例。结果流式细胞仪分析细胞显示CD29、CD44、CD105表达阳性,CD45、CD14、CD34表达阴性,符合BM-SCs的特点。谷氨酸诱导组较多细胞呈凋亡改变,BMSCs的凋亡率较对照组升高(P〈0.05),且随着谷氨酸浓度的增加,细胞凋亡率也相应增加。结论谷氨酸可诱导BMSCs毒性损伤。

Abstract:Objective To explore the role of glutamate in inducing damage of rat bone marrow mesenchymal stem cells (BMSCs). Methods Primary cultured rat BMSCs were incubated. Cell phenotype was identified by flow cytometry. Apoptosis was detected using 4' ,6-diamidino-2-phenylindole(DAPI) fluorescence staining. BMSCs survival rate was measured by MTT assay, Apoptotic cells were counted by flow eytometry following PI/Annexin-V staining. Results Cell phenotype identification showed positive antigen expressions with CD29, CD44 , CD105 and negative antigen expressions with CD43, CD14, CD34. Some of the cells showed apoptosis changes in glutamate induced group. In comparison to the control group, the apoptotic rate of glutamate induced group increased ( P 〈 0. 05 ), and it increased in proportion to the increase of glutamate concentration. Conclusions Glutamate could induce BMSCs damage.
