摘    要:目的:在人胎儿脑组织cDNA中钓取并克隆新基因.方法:以RT-PCR方法用人FGF-19特异性引物从人胎脑cDNA中扩增到一大小约660bp的片段,并连入TA克隆载体测序,得到一个表达序列标签(EST),输入GeneBank、Swissprot、dbEST等数据库中进行同源比较分析.结果:分析表明它是人胎儿脑组织未知基因EST,是一个663个碱基可编码220个氨基酸的开放读码框架.结论:在人胎儿脑组织cDNA中钓取并克隆一个人胎脑新基因EST.

Abstract:Objective:To obtain and clone the new gene of human fetal brain. Methods: A cDNA fragment about 660 bp was isolated from human fetal brain tissue by RT PCR with human FgF 19 primers. Then it was cloned into pGEM T Easy plasmid to be sequenced. Results: Sequencing analysis proved that it was a novel Expressed Sequence Tag(EST) derived form human fetal brain and contained an open reading frame(ORF) with the size of 663 bp coding for 220aa.Conclusion: A human novel EST was obtained.
