金昭胶囊对急性酒精性肝损伤小鼠的保护作用

摘    要:目的:研究金昭胶囊对急性酒精性肝损伤的保护作用机制,为其临床应用提供实验依据。方法:将美国癌症研究所(ICR)小鼠随机分为空白组、模型组、阳性药组、金昭胶囊低、中、高剂量组,每组10只,持续给药4周后,用50%乙醇灌胃制备小鼠急性酒精性肝损伤模型。观察各组小鼠状态,测定醉酒状态、血清转氨酶活性,肝组织乙醇及脂肪代谢过程相关酶活性、抗氧化酶活性、胃组织抗氧化酶活性并取肝脏进行苏木精-伊红(HE)染色病理切片检查。结果:与模型组比较,金昭胶囊各剂量组小鼠血清中谷丙转氨酶(GPT)、谷草转氨酶(GOT)活性显著降低(P<0.01,P<0.05);肝组织中总胆固醇(TC)、三酰甘油(TG)、丙二醛(MDA)活性显著降低(P<0.01,P<0.05),超氧化物歧化酶(SOD)、过氧化氢酶(CAT)、还原型谷胱甘肽(GSH)、乙醇脱氢酶(ADH)、乙醛脱氢酶(ALDH)活性显著升高(P<0.01,P<0.05),肝细胞坏死程度降低;胃组织丙二醛(MDA)活性显著降低(P<0.01),超氧化物歧化酶(SOD)、一氧化氮(NO)、前列腺素E2

Protective Effect of Jinzhao Capsules on Acute Alcoholic Liver Injury in Mice

Abstract:To study the mechanism of Jinzhao Capsules in the protection against acute alcoholic liver injury and provide experimental evidence for the clinical application of the capsules.Methods:The Institute of Cancer Research(ICR) mice were randomly assigned into a blank group,a model group,a positive drug group,and low-,medium-,and high-dose Jinzhao Capsules groups,with 10 mice in each group.After 4 weeks of administration,50% ethanol was used to establish the mouse model of acute alcoholic liver injury.The state of mice was observed,and the drunken state,serum transaminase activity,activities of the enzymes associated with liver tissue ethanol and fat metabolism,and activities of antioxidant enzymes in the liver tissue and stomach tissue were determined.The liver samples were collected for pathological examination via hematoxylin-eosin(HE) staining.Results:Compared with the model group,Jinzhao Capsules weakened the activities of glutamic-pyruvic transaminase(GPT) and glutamic-oxaloacetic transaminase(GOT) in the serum(P<0.01,P<0.05),lowered the levels of total cholesterol(TC),triacylglycerol(TG),and malondialdehyde(MDA) in the liver tissue(P<0.01,P<0.05),increased the activities of superoxide dismutase(SOD),catalase(CAT),glutathione(GSH),alcohol dehydrogenase(ADH),and aldehyde dehydrogenase(ALDH)(P<0.01,P<0.05),and reduced the liver cell necrosis.Furthermore,the capsules lowered the level of MDA in stomach tissue(P<0.01) and elevated the levels of SOD,nitric oxide(NO),and prostaglandin E2(PGE2)(P<0.01,P<0.05).Conclusion:Jinzhao Capsules may protect mice from acute alcoholic liver injury by enhancing the activity of key enzymes in alcohol metabolism and antioxidant enzymes in the liver tissue,inhibiting the production of lipid peroxides,protecting the integrity of liver cell membrane,and reducing the release of transaminase.At the same time,Jinzhao Capsules can protect the gastric mucosa by regulating the activity of peroxidase in the gastric tissue.
