缬沙坦联合还原型谷胱甘肽对糖尿病肾病的保护作用

目的探讨缬沙坦及还原型谷胱甘肽对糖尿病肾病大鼠保护作用及可能机制。方法以链脲佐菌素诱导糖尿病大鼠模型,同时设对照组、模型组、缬沙坦干预组、还原型谷胱甘肽干预组、缬沙坦联合还原型谷胱甘肽干预组,连续治疗8周后,HE染色检测肾组织病理变化;腹主动脉采血行血液动力学检测,以酶联免疫吸附测定法行血管紧张素Ⅱ的测定。结果与模型组相比缬沙坦和还原型谷胱甘肽均能改善糖尿病肾病大鼠的血液动力学指标(P<0.05),Val组和Val&GSH组血浆中AngⅡ升高(P<0.05)。结论缬沙坦联合还原型谷胱甘肽可加强机体高糖环境下抗氧化作用,减少糖尿病肾病大鼠机体内肾素血管紧张素系统(RAS)对肾脏的损伤,改善血液黏稠度作用,在一定的程度上可以延缓糖尿病肾病的进展。

The Protect Effect of Combining Valsartan and Reduced Glutathione to Diabetic Nephropathy

Objective To investigate the protection function and the possible mechanism of combining valsartan and reduced glutathione to dabetic nephropathy rats.Methods Streptozotocin(STZ) induced diabetic rats were randomly divided into five group: control group,diabetic nephropath group(DN),valsartan treatment group(Val),reduced glutathione treatment group(GSH) and combine valsartan with reduced glutathione treatment group(Val&GSH).All the groups accpeted continuous treatment for eight weeks,The renal pathological changes were evaluated by HE staining;The blood was collected from the abdominal aorta to examine the indexes of hemorheology;Enzyme-linked immunosorbent assay was used to detect serum angⅡ.Results Compared with DN group,the indexes of hemorheology was improved in all treatment group(P<0.05),the levels of serum angⅡwere higher(P<0.05) in Val group and Val&GSH group.Conclusion In the condition of high sugar valsartan combined with glutathione can strengthen the oxidation resistance of diabetic nephropathy rats,reduce the renal injury which was induced by RAS,also the two drugs can Improve the hemorheology indexes and In a certain extent delay the progress of diabetic nephropathy.
