参附注射液预处理对缺血再灌注损伤保护作用的影响

摘    要:目的探讨参附注射液预处理对心肌缺血再灌注损伤的保护作用及机制。方法新西兰大白兔30只,随机分成2组:缺血再灌注损伤组(IR)、参附注射液预处理组(SFI)。实验期间描记心功能指标:左室内压力峰值(LVSP)、左室舒张末期压(LVEDP)、左室内压变化最大速率(±dp/dtmax)。2组均于再灌注30min后立即取结扎线下约0.5cm处心肌组织1g,制成组织匀浆,采用高速离心法制备线粒体悬液,检测琥珀酸脱氢酶(SDH)、谷胱甘肽过氧化物酶(GSH-PX)的活力及丙二醛(MDA)含量。结果与IR组比较,SFI组各时点的收缩及舒张功能下降程度明显减轻(P<0.05),MDA含量显著降低(P<0.05),GSH-PX活力显著升高(P<0.05),SDH活力明显升高(P<0.01)。结论参附注射液预处理能较明显地抑制缺血再灌注损伤时线粒体脂质过氧化作用,降低MDA的含量,提高GSH-PX和SDH的活力,保护心肌细胞线粒体的结构和功能,对再灌注引起的损伤起到保护作用。

Abstract:Objective To investigate the protective effects and its mechanism of Shenfu injection preconditioning on myocardial ischemia-reperfusion injury.Methods 30 New Zealand white rabbits, were randomly divided into two groups:ischemia-reperfusion injury group(IR,n=15),Shenfu injection preconditioning group(SFI,n=15). The model of myocardial ischemia-reperfusion injury was produced by coronary artery ligation. The systolic and diastolic function of left ventricle including LVSP,LVEDP and ±dp/dt_ max was measured. The activities of SDH,GSH-PX,MDA were observed respectively.Results As compared with those in IR group, the systolic and diastolic function of left ventricle in SFI group was obviously decreased at different time point (P<0.05).The activities of SDH,GSH-PX were significantly increased (P<0.05 and P<0.01 respectively),MDA content was notably decreased (P<0.05).Conclusion Shenfu injection has obvious protective effects on myocardial ischemia-reperfusion injury,which could decrease MDA content,increase the activities of GSH-PX and SDH,inhibit the effects of mitochondria lipid proxidation and protect the structure and function of mitochondria in myocardial cells.
