子宫内膜腺上皮细胞和基质细胞的分离培养

摘    要:目的 建立一种获得高产量子宫内膜腺上皮细胞和基质细胞的分离培养方法。方法 通过胶原酶消化法、系列过滤法及贴壁纯化等技术分离纯化培养人子宫内膜腺上皮细胞和基质细胞 ,并通过光镜及免疫细胞化学染色进行鉴定。结果 腺上皮细胞呈蝌蚪形 ,漩涡状生长。角蛋白免疫细胞化学染色阳性 ,纯度约 90 %。基质细胞呈梭形或多角形 ,平行状生长。波形蛋白染色反应阳性 ,纯度达 95 %。每例子宫肌瘤切除标本可获得 (10~2 5 )× 10 6 原代基质细胞和 (4~ 6 )× 10 6 原代腺上皮细胞 ,每例诊断性刮宫获得的原代细胞数约为前者的 1/ 2~2 / 3。结论 改良培养程序可获得高产量的纯化的子宫内膜腺上皮细胞和基质细胞。

Isolation and culture of human epithelial and stromal cells from endometrium
Abstract:Objective To establish a method of isolation and culture to harvest a better yield of human epithelial and stromal cells.Methods Enzymolysis,serial filtration and adhesion purification were used to isolate,purify human epithelial and stromal cells.These two types of cells were labeled by light microscopy and immunocytochemical staining.Results Epithelial cells had tadpole shape and grew in whorls.They were stained positively by CK19 (cytokeratin 19) with 90%.Stromal cells were fusiform or polygonal-shaped.They were stained positively by vimentin about 95%.The yields of primary culture of stromal cells and epithelial cells were about (10~25)×106 and (4~6)×106 respectively from one endometrium sample undergoing hysteromyomectomy.The yields of primary cells from endometrium by curettage were about 1/2~2/3 of the former.Conclusion A good yield of purified human epithelial and stromal cells can be obtained by the modified culture procedure.
