肿瘤疫苗对小鼠肝癌作用的研究

摘    要:目的 :通过动物实验评价肿瘤疫苗对肝癌的防治作用 ,为临床应用提供实验依据。方法 :用小鼠肝癌细胞系MM45T .Li经过化学修饰制成肿瘤疫苗免疫小鼠 ,然后再用该细胞系接种于小鼠腋下。分别观察免疫鼠及未免疫小鼠的肿瘤生长情况 ,进行病理学检查 ;并用3H -TdR释放法检测小鼠脾细胞对MM45T .Li细胞的杀伤能力差异。结果 :①经肿瘤疫苗免疫的小鼠的肿瘤生长速度明显低于未免疫小鼠 ;②病理学检查发现免疫鼠肿瘤组织内大量淋巴细胞浸润 ,未免疫鼠内罕见淋巴细胞浸润 ;③3H -TdR释放实验显示免疫鼠脾细胞对MM45T .Li细胞的杀伤能力明显高于未免疫鼠。结论 :按我们现行方法制备的肿瘤疫苗能激活小鼠抗肝癌的免疫反应 ,抑制肝癌的生长 ,但完全保护作用尚不明确

Abstract:Objective To evaluate the role of the tumor vaccine which was got by chemically treating MM45T.Li,the mice Hepatocellular cell line,to protect against the hepatocellular carcinoma by animal test and to provide an experimental basis for clinical application.Methods In each mice groups including immunized and unimmunized,subcutaneouslly injecting 2×10 6 MM45T.Li cells to make the tumor grow.Then observation was made on the growth of tumors in defferent groups,carrying on pathological examination and cytotoxity of the mice spleen cells to the MM45T.Li cells by 3H-TdR-release test.Results The tumor growth rates of the immunized group were much lower than those of the unimmunized group;The infiltrated lymphocytes in tumors were generally seen in the immunized group but,rare in the unimmunized group;The ability of the spleen cell cytotoxity of immunized group were much stronger than that of the unimmunized group.Conclusion Mice could obtain protection against the hepatocellular carcinoma by injection of the tumor vaccine prepared by us,but how it works is still unknown.
