重庆市地产药材蓼子草质量标准研究

摘    要:目的建立重庆市地产药材蓼子草的质量标准。方法采用显微镜观察蓼子草粉末的纤维特征,采用薄层色谱法定性鉴别蓼子草中的成分金丝桃苷;采用高效液相色谱法测定槲皮素的含量,色谱柱为AgilentEclipseXDB-C18柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.4磷酸水溶液(55∶45,V/V),流速为1.0 mL/min,检测波长为360 nm,柱温为30℃,进样量为10μL。结果蓼子草显微特征包括气孔、非腺毛、花粉粒、草酸钙簇晶体;蓼子草薄层色谱图中金丝桃苷特征明显,专属性强;槲皮素的质量浓度在1.42~71.20μg/mL范围内与峰面积线性关系良好(r=0.9999,n=5),精密度、稳定性、重复性试验的RSD均小于2.0%(n=6),平均加样回收率为98.00%,RSD为0.81%(n=6)。结论该方法操作简便,结果准确,专属性强,稳定性和重复性均较好,可用于蓼子草的质量控制。

Abstract:Objective To establish a quality standard of Chongqing native herb Polygonum criopolitanum.Methods The fiber characteristics of Polygonum criopolitanum powder were observed by microscope,and the hyperoside in Polygonum criopolitanum powder was qualitatively identified by the thin-layer chromatography(TLC)method.The content of quercetin was determined by the high-performance liquid chromatography(HPLC)method.The chromatographic column was Agilent Eclipse XDB-C18 column(250 mm×4.6 mm,5μm),the mobile phase was methanol 0.4 phosphoric acid aqueous solution(55∶45,V/V),the flow rate was 1.0 mL/min,the detection wavelength was 360 nm,the column temperature was 30℃,and the injection volume was 10μL.Results The microscopic characteristics of Polygonum criopolitanum included stomata,non-glandular hairs,pollen grains and calcium oxalate cluster crystals.Hyperoside in the TLC chromatogram of Polygonum criopolitanum had obvious characteristics and strong specificity.The linear range of quercetin was 1.42-71.20μg/mL(r=0.9999,n=5).The RSDs of precision,stability and repeatability tests were less than 2.0%(n=6).The average recovery of quercetin was 98.00%with an RSD of 0.81%(n=6).Conclusion The method is simple,accurate,specific,stable and reproducible,which can be used for the quality control of Polygonum criopolitanum.
