大豆皂甙对人肺腺癌细胞增殖抑制作用研究

摘    要:目的 探讨大豆皂甙 (total soysaponin,TS)对体外培养人肺腺癌细胞系 SPC- A- 1的直接抑制作用及可能机制。方法 通过克隆形成法、3H- Td R掺入法与四氮唑蓝比色法观察不同浓度 TS的抑瘤效应。结果 给药后 SPC- A- 1细胞增殖受抑制 ,0 .5 mg/ ml剂量组抑制作用最佳 ,并出现细胞脱落、胞浆内颗粒状物堆积等形态学改变 ;动态观察发现给药后 2 4 h已有明显抑制作用 ,72 h达高峰。给药组克隆形成数、cpm和 A值明显少于对照组 (P<0 .0 5) ,3H- Td R掺入抑制率增加 ,细胞生存率下降。结论  TS通过抑制细胞 ;DNA合成、干扰细胞代谢及改变细胞外衣的性质 ,抑制贴附型细胞贴壁生长 ,对 SPC- A- 1细胞具有剂量依赖和时间依赖方式的抑制作用。

Abstract:Objective To investigate the direct inhibitory effects of total soysaponin(TS) on the growth of cultured human adenocarcinoma cells of lung SPC-A-1 and possible mechanism.Methods The methods of cell counting, colony forming assay, tetrazolium (MTT) colorimetric assay and 3H-TdR incorporation assay were used to detect the inhibitory effects of TS with different dosages.Results TS could inhibit proliferation of SPC-A-1 cells, the best concentration was 0.5mg/ml. The inhibitory effect occurred at 24 hour significantly, the strongest inhibition occurred at 72 hour. At the same time, some morphological changes such as appearance of granular bodies in plasma and detachment from wall could be observed. The numbers of colony formation,A value and cpm value reduced obviously, the inhibitory rate of 3H-TDR incorporation increased and the cell survival rate in TS group also decreased compared to the control group.Conclusions TS could inhibit the proliferation of adenocarcinoma cells in a dose-dependent manner and time-dependent manner by blocking DNA synthesis, obstructing the metabolism of cells, changing the characteristics of cell coat and finally inhibiting the adhesion to wall of adenocarcinoma cells.
