姜黄素抑制宫颈癌细胞株增殖和诱导凋亡的体外研究

摘    要:目的:探讨中药姜黄素(Cur)对子宫颈癌SiHa细胞株的增殖抑制和诱导凋亡的作用。方法:以不同浓度的姜黄素(10umol/L、20umol/L、30umol/L),分12h、24h、48h、72h4个时间点处理SiHa细胞,光镜观察细胞形态变化;用四甲基偶氮唑蓝(MTT)法测定细胞增殖抑制率;用透射电子显微镜及琼脂糖凝胶电泳检测凋亡的发生。结果:姜黄素可抑制SiHa细胞增殖,作用呈明显的时效和量效关系,差异有非常显著性(P〈0.01);姜黄素可诱导SiHa细胞凋亡,透射电子显微镜下可见凋亡细胞;琼脂糖凝胶电泳出现细胞凋亡典型的DNA“梯状”条带。结论:姜黄素体外对SiHa细胞具有增殖抑制作用和促进凋亡作用。

Abstract:Objective:To investigate the SiHa cells growth inhibition and Apoptosis induced by Curcumin.
