高压氧对一氧化碳中毒大鼠海马神经细胞线粒体膜电位与细胞凋亡的影响

摘    要:目的 观察一氧化碳中毒(COP)后脑损伤时线粒体膜电位及细胞凋亡数量的变化以及高压氧(HBO)对其影响。方法Wistar大鼠共128只,采用配伍组设计,按随机抽样原则将动物分成4组16小组。正常对照组8只;CO组COP后第1,5,10,15,20天各8只;CO+HP组高气压处理后第1,5,10,15,20天各8只:CO+HBO组HBO处理后第1,5,10,15,20天各8只。用流式细胞仪测定COP大鼠在不同时间段海马神经细胞线粒体膜电位及海马神经细胞凋亡细胞相对百分比(%)的变化和HBO治疗后的改变。结果CO组在COP后大鼠海马神经细胞线粒体膜电位在第1,5,10天降低(P<0.01-0.05);CO+HBO组在第1,5天降低(P<0.01-0.05)。CO组COP后大鼠海马神经细胞凋亡细胞相对百分比在第1,5,10,15,20天高于对照组(P<0.01);而CO+HBO组只在第1,5,10天高于对照组(P<0.01)。结论COP后大鼠海马神经细胞线粒体膜电位降低,凋亡细胞数增加,并持续很长时间,而HBO处理可缩短线粒体膜电位降低的减少凋亡细胞数百分比。

Abstract:Objective To study the changes of mitochondrial membrane potential (MMP) and percentage of apoptotic neural cells in rats with brain injury after carbon monoxide poisoning and to e- valuate the effect of hyperbaric oxygen treatment. Methods 128 Wistar rats were randomly divided in- to 16 groups with each group 8 animals, namely: 5 CO groups (carbon monoxide exposure ), 5 CO HP groups (carbon monoxide exposure and high atmospheric pressure treatment ), 5 CO HBO groups (car- bon monoxide exposure and hyperbaric oxygen treatment ) and 1 control group (no carbon monoxide ex- posure and no hyperbaric oxygen treatment ). All the rats except that of the control group were exposed to 3 378. 32mg/L CO for 1 hour. Then the rats of CO HP groups and CO HBO groups were given high atmospheric pressure treatment and hyperbaric oxygen treatment after the CO exposure. Using flow cytometry, mitochondrial membrane potential and the percentage of apoptotic neurons were mea- sured. Results MMP of neuron was significantly lower in CO groups than in control group at 1st, 5th and 10th day after carbon monoxide poisoning; MMP was also lower in CO HP groups at 1st, 5th, 10th and 15th day, and in CO HBO groups at 1st and 5th day. At the same time, the percentages of apop- totic cells of the hippocampus in CO groups were significantly increased as compared with the control af- ter CO exposure at 1st, 5th, 10th, 15th and 20th day; whereas they were increased at 1st, 5th and 10th day only in CO HBO groups. No notable differences were observed between CO and CO HP groups. Conclusions After carbon monoxide poisoning, the mitochondrial membrane potential of neuron was decreased and the percentage of apoptotic neurons was increased as the signs of brain injury, and these changes could continue for 15-20d or longer; but with hyperbaric oxygen treatment, they were signifi- cantly recovered within 10-15 days.
