罗格列酮对2型糖尿病大鼠白内障治疗作用的实验研究

摘    要:目的 探讨罗格列酮对2型糖尿病大鼠并发白内障的治疗作用。方法 采用高脂高糖饲料喂养及小剂量注射链脲佐菌素(streptozocin,STZ)的方法,制备2型糖尿病性白内障大鼠模型;采用罗格列酮干预治疗,定期监测血糖和观察晶状体的变化。采用末端脱氧核苷酸转移酶介导的 dUTP缺口标记法(terminal deoxynucleotidyltransferase mediated dUTP nick end labeling,TUNEL)标记凋亡的晶状体上皮细胞(lens epithelial cells, LECs),测定凋亡百分率;测定血浆胰岛素和血脂水平。结果 TUNEL染色显示模型组大鼠 LECs中有凋亡阳性细胞,糖尿病性白内障组与同期正常对照组比较,其LECs凋亡百分率显著升高(P<0.05),且随时间的推移、晶状体混浊程度加重,凋亡率呈明显上升趋势;罗格列酮治疗组凋亡率明显低于未治疗组(P<0.05)。结论 罗格列酮降低血糖的机制不是通过刺激胰岛素分泌而是增加机体对胰岛素的敏感性,对糖尿病性白内障有明显的治疗作用。

Abstract:ObjectiveTo investigate the therapeutic effect of rosiglitazone on type 2 diabetic rats with subsequent cataract. MethodsModels of type 2 diabetic cataract rats were established by feeding high fat and high glucose food and injecting small dosage streptozocin (STZ). Model rats were treated by rosiglitazone and their blood glucose and changes of lens were monitored during period of experiment. Terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling(TUNEL) staining was used to monitor the apoptosis of lens epithelial cells(LECs) and determine the percentage of apoptoic cells. The levels of plasm insulin and blood lipid were measured. ResultsCompared with control group in the corresponding period , the percentage of apoptotic cells in model group increased significantly(P<(0.05)), furthermore, along with the time going on and the degree of lens opacity increasing, the percentage of apoptotic cells increased obviously. The percentage of apoptotic cells in rosiglitazone group was significantly lower than that of the untreated group(P<(0.05)). ConclusionRosiglitazone can depress blood glucose, which may result from the increasing sensibitity to insulin but not from stimulating insulin excretion. Rosiglitazone has significant therapeutic effect on diabetic cataract.
