RP-HPLC法测定大豆提取物中大豆苷元、染料木素、大豆苷、染料木苷的含量

目的：建立ＲＰ－ＨＰＬＣ法分离检测大豆提取物大豆苷元，染料木素、大豆苷、染料木苷４成分含量的方法，方法：用ＺorbaxSB-C18色谱柱，以甲醇－乙腈－０．１％，磷酸（３０：８：６２）和甲醇－乙腈－０．１％磷酸（１６：２４：６０）为流动相分别测定大豆苷元，染料木素和大豆苷、染料木苷、检测波长为２５４nm.结果：大豆苷元、染料木素、大豆苷、染料木苷４成分的平均回 分别为９８．７％，９９．５％，９８．４％，９８．６５，ＲＳＤ分别为１．４％，０．９％，０．９％，１．７％（n=４）。结论方法简便、准确、重复性好。

RP-HPLC determination of daidzein, genistein, daidzin and genistin in soybean extract

Object A method for the determination of daidzein, g enistein,daidzin and genistin in soybean extract by RP-HPLC was established . Methods Zorbax SB-C18 column (4.6×15 mm, 5 μm) was us ed . Daidzein and genistein were separated with mobile phase Ⅰ∶methanol-acetoni tr ile-0.1% phosphoric acid solution (30∶8∶62). Daidzin and genistin were sepa rate d with mobile phase Ⅱ∶methanol-acetonitrie-0.1% phosphoric acid solution ( 16∶ 24∶60). The UV detection wavelength was 254 nm. Results The ave rage recoveries of daidzein, genistein, daidzin and genistin were 98.7%, 99.5 %, 98.4%, 98.6% and the precision (RSD) were 1.4%, 0.9%, 0. 9%, 1.7% ( WTBX n=4) respectively. Conclusion: The method was ac curate, rapid and with good reproducibility.
