摘    要:目的分析内源性端粒酶抑制基因PinX1在膀胱癌中的表达,探讨PinX1在膀胱癌发生、发展和预后中的作用及临床意义。方法应用半定量反转录聚合酶链反应(RT-PCR)检测48例膀胱癌组织中内源性端粒酶抑制基因PinX1及端粒酶反转录酶hTERT的mRNA表达水平,同时应用免疫组织化学方法检测PinX1蛋白表达水平。结果PinX1表达与膀胱癌组织分化、临床分期和淋巴转移之间呈显著相关性,与hTERT表达呈负相关。结论PinX1的下调可能与膀胱癌形成、转移过程中端粒酶激活及维持机制有关。检测PinX1的表达水平对评价膀胱癌恶性程度、转移潜能和预后评估均有重要的临床价值。

Abstract:Objective To investigate the role of endogenous telomerase catalytic inhibitor PinX1 in the progression of bladder cancer and its clinical significance.Methods The tissue samples containing 48 cases of bladder cancer were studied.The expression of PinX1,telomerase subunits(hTERT) were detected by semi-quantitative RT-PCR.Immunohistochemistry methods were used to examine the protein expression of PinX1 gene.Results PinX1 expression is significantly correlated with the histological differentiation,clinical stage and lymph node metastasis of bladder cancer.The level of PinX1 is negatively associated with the expression of hTERT.Conclusion The down-regulation of PinX1 may function as a telomerase activation factor and thus play an important role in the clinical phenotype and development of bladder cancer.PinX1 gene may be used as one of the molecular markers to determine the malignant degree,metastasis potency and predicting progression of bladder cancer.
