糖尿病大鼠肾皮质糖基化终产物含量与纤溶酶原激活物抑制物-1表达的关系

摘    要:目的:研究链脲佐菌素诱发的糖尿病(DM)大鼠肾皮质糖基化终产物(AGEs)含量与纤溶酶原激活物抑制物-1(PAI-1)表达的关系及牛磺酸的调节作用.方法:DM大鼠用牛磺酸处理14周后,分别用荧光法、生物化学法和Northern杂交检测肾皮质AGEs含量、组织型纤溶酶原激活物(t-PA)活性、PAI-1活性和PAI-1 mRNA表达.结果:与对照大鼠相比,DM大鼠肾皮质AGEs含量、PAI-1活性和PAI-1 mRNA表达明显增加,而t-PA活性下降.AGEs含量与PAI-1生物活性及PAI-1 mRNA表达均呈正相关(P<0.01).DM大鼠给予牛磺酸处理后可改善上述参数的改变.结论:DM大鼠肾皮质AGEs含量增加能明显上调PAI-1基因表达,牛磺酸可抑制AGEs形成和PAI-1基因表达的上调.

Abstract:Objective:To investigate the relationship between the advanced glycation end products(AGEs) content and plasminogen activator inhibitor 1 (PAI 1) gene expression in renal cortex of streptozotocin induced diabetic rats and its modulation by taurine. Methods:The fluorescence method, biochemical technique and Northern blot analysis were used to detect the AGE content, tissue plasminogen activator(t PA)activity,PAI 1 activity and PAI 1 mRNA expression, respectively. Results:Compared with control animals, diabetic rats showed an increase in AGE content,PAI 1 activity and PAI 1 mRNA expression,and a decrease in t PA activity in renal cortex. AGE content was positively correlated with PAI 1 activity and PAI 1mRNA expression. Administration of taurine to diabetic rats improved the changes of these parameters. Conclusion:The increased AGE content in renal cortex of diabetic rats can markedly upregulate PAI 1 mRNA expression, and taurine suppresses AGEs formation and PAI 1 mRNA expression.
