金茵清热口服液对PBMC中TLRs mRNA表达的影响

摘    要:目的:研究金茵清热口服液(试药)对人外周血单个核细胞(PBMC)中Toll样受体(TLRs)mRNA水平的影响,探索试药在TLRs信号通路中的免疫调节作用。方法:体外分离、培养人PBMC,设试药组和空白对照组,试药组加入试药刺激PBMC,与空白组经37℃,5% CO2培养24 h后,用Trizol抽提PBMC总RNA,采用实时荧光定量PCR(qRT-PCR)法检测PBMC中TLR1-10 mRNA水平。结果:与空白对照组比较,试药组PBMC中,除TLR5、TLR7、TLR8、TLR10 mRNA水平下调(aP<0.05,cP>0.05),其他TLRs mRNA表达均上调,其中TLR1、TLR2和TLR6上调最为显著,分别上调4.11,6.32和5.58倍(bP<0.01)。结论:金茵清热口服液可能主要通过影响TLR1、TLR2和TLR6来激活天然免疫应答,发挥免疫调节作用。

Abstract:OBJECTIVE To study the effect of Jinyin Qingre Oral Liquid on the mRNA expression of Toll-like receptors (TLRs) in human peripheral blood mononuclear cells (PBMC) and explore the immune regulation effects of Jinyin Qingre Oral Liquid in the TLRs signaling pathways. METHODS Human PBMCs were isolated and cultured in vitro. The test drug group and the blank control group were assigned. The PBMCs in test drug group were stimulated by Jinyin Qingre Oral Liquid and incubated with the blank control group at 37℃, 5%CO2 for 24 h. Total RNA in PBMCs was extracted with Trizol, and the levels of TLR1-10 mRNA were detected with real-time fluorescence quantitative PCR (qRT-PCR) method. RESULTS Compared with the blank control group, Jinyin Qingre Oral Liquid treatment led to significant increase in the mRNA expression levels of TLR1, TLR2 and TLR6 by 4.11, 6.32 and 5.58 folds (bP<0.01), respectively, while on the other hand led to decrease in the mRNA expression levels of TLR5, TLR7, TLR8 and TLR10 (aP<0.05,cP>0.05). CONCLUSION Jinyin Qingre Oral Liquid may activate the innate immune response to exert immune regulatory effect through up-regulating TLR1, TLR2 and TLR6.
