基于网络药理学的复方当归注射液治疗缺血性脑损伤作用机制分析

摘    要:目的:采用网络药理学的方法探讨复方当归注射液治疗缺血性脑损伤的作用机制。方法:运用中药系统药理学分析平台(TCMSP)检索复方当归注射液3味中药化学成分和相关作用靶点。利用Cytoscape 3.7.0构建化合物-靶点网络,采用STINGV10.5软件构建蛋白互作PPI网络,在此基础上进行基因本体GO和京都基因与基因组百科全书KEGG生物通路富集分析。结果:从复方当归注射液3味中药中筛选得到24个活性成分,其作用于99个靶点,涉及的关键靶点包括AKT1、JUN、ESR1、IL6、EGF、F2、TNF和PIK3CG等;调节信号通路包括神经活性受体-配体相互作用通路、神经激素信号通路、MAPK信号通路、cGMP-PKG信号通路、多巴胺能突触、VEGF信号通路、血小板激活和PPAR信号通路;参与含氧化合物的反应、有机环状化合物反应、内生刺激的反应、细胞内信号传导的正调节和活性氧物质代谢过程的调节等多种生物过程。结论:本研究初步阐明了复方当归注射液抗缺血性脑损伤的药理作用及其相关机制,为进一步深入研究奠定良好基础,为研制抗缺血性脑损伤新药提供参考依据。

Abstract:OBJECTIVE To investigate the mechanism of compound angelica injection in the treatment of ischemic brain damage by using network pharmacology. METHODS Chemical components and interactive targets related to three traditional Chinese medicine (TCM) herbs were identified by the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP). Compound-target network was constructed using Cytoscape 3.7.0, and protein interaction PPI network was constructed using STINGV10.5 software, based on which Gene Ontology GO and Kyoto Encyclopedia of Genes and Genomes KEGG biological pathway enrichment analysis were performed. RESULTS A total of 24 active ingredients were screened from 3 kinds of traditional Chinese medicines, which acted on 99 targets. The key targets included AKT1, JUN, ESR1, IL6, EGF, F2, TNF and PIK3CG. The signal pathways included neuroactive receptor-ligand interaction pathway, neurohormone signaling pathway, MAPK signaling pathway, cGMP-PKG signaling pathway, dopaminergic synapse, VEGF signaling pathway, platelet activation and PPAR signaling pathway. They were involved in many biological processes, such as the reaction of oxygen-containing compounds, the reaction of organic cyclic compounds, the reaction of endogenous stimulation, the positive regulation of intracellular signal transduction and the regulation of metabolic process of active oxygen CONCLUSION S In this study, the pharmacological effects of compound angelica injection against ischemic brain injury and its related mechanisms were preliminarily elucidated, which laid a good foundation for further research and provided a reference basis for the development of new drugs against ischemic brain injury.
