摘    要:目的:探讨莪术油注射液对宫颈癌细胞Hela增殖的抑制作用及潜在作用机制。方法:采用CCK-8法筛选莪术油最佳给药剂量,并设立对照组和莪术油处理高、中、低剂量组。CCK-8法检测莪术油对Hela细胞生长曲线的影响;流式细胞术检测莪术油对Hela细胞周期的影响;Western Blot试验检测莪术油对Hela细胞中Wnt3a、β-catenin、C-myc、CyclinD1蛋白表达的调节作用。结果:CCK-8试验表明,莪术油给药剂量越大,其对Hela细胞增殖的抑制作用越明显,以高剂量组为最优。流式细胞术试验表明,莪术油可将Hela细胞阻滞在G1期,使S期和G2期细胞比例明显降低。Western Blot试验结果显示,莪术油处理后Wnt3a、β-catenin、C-myc、CyclinD1蛋白表达相较于对照组其表达明显降低。结论:莪术油注射液通过调节Wnt/β-catenin信号通路抑制宫颈癌细胞增殖。

Abstract:OBJECTIVE To investigate the inhibitory effect of zedoary turmeric oil injection on the proliferation of cervical cancer cell line Hela and its potential mechanism.METHODS The optimal dose of zedoary turmeric oil was screened by CCK-8 method,and the control group and zedoary turmeric oil were used to treat high,medium and low dose groups.CCK-8 assay was used to detect the effect of zedoary turmeric oil on the growth curve of HeLa cells;flow cytometry was used to detect the effect of zedoary turmeric oil on the cell cycle of HeLa cells;Western blot assay was used to detect the regulatory effect of zedoary turmeric oil on the expression of Wnt3a,β-catenin,C-myc,and CyclinD1 protein in HeLa cells..RESULTS The CCK-8 test showed that higher dose of zedoary turmeric oil showed more inhibitory effect on Hela cell proliferation with high dose group being the best.Flow cytometry assay showed that zedoary turmeric oil could arrest HeLa cells in G1 phase,which significantly reduced the proportion of cells in S phase and G2 phase.Western Blot test results showed that the expression of Wnt3a,β-catenin,C-myc,and CyclinD1 protein was significantly reduced after treatment with zedoary turmeric oil compared with the control group.CONCLUSION Zedoary turmeric oil injection can inhibit the proliferation of cervical cancer cells by regulating Wnt/β-catenin signaling pathway.
