顺铂对肝癌细胞凋亡及其细胞周期的影响

摘    要:目的:探讨顺铂对Hca-F细胞凋亡及细胞周期的影响。方法:采用荧光,透射电镜及流式细胞术分析法检测肝癌细胞凋亡,细胞周期和增殖指数(PI)的变化。结果:CDDP可引起肿瘤细胞凋亡,并随着给药时间的延长,药物浓度的增高,细胞凋亡率随之增高,细胞凋亡形态结构越发典型,表现为核固缩,染色质边集,核小体形成等,同时药物浓度的升高,细胞周期S期比例下降,G0/G1期比例上升,PI增殖指数下降。结论:CDDP可引起细胞凋亡,细胞周期G0/G1期阻滞,使细胞分裂增殖活动受到抑制。

Abstract:Objective:To observe the correlation between CDDP and Hca F cell lines in the qualitative and quantitative of apoptosis,cell cycle and proliferation index.Methods:The staining of Hoechst 33342 and PI fluorecence,transmission electron microscopic technique and flow cytometry(FCM)were used to detect the above results.Results:CDDP could induce Hca F cell apoptosis markedly.The ratio of apoptosis would increase with time lengthening and the concentration of CDDP,and would appear PI and S phase decreasing as well as G 0/G 1 phase increasing.Conclusion:CDDP can induce Hca F cell apoptosis,arrest cell cycle and inhibit cell proliferation.
