大黄素对人胃癌细胞MKN45增殖抑制及诱导凋亡的研究

目的：研究大黄素（Emodin）对人胃癌细胞MKN45增殖及诱导凋亡的作用。方法：用噻唑蓝（MTT）法观察Emodin对MKN45的生长抑制作用;用DNA Ladder凝胶电泳法、荧光染色法、流式细胞仪法分析Emodin的诱导凋亡作用。结果：与对照组相比,Emodin能明显抑制MKN45细胞的生长且与浓度、时间呈依赖性,其IC50（48小时）为（47.5±0.3）μmol/L;Emodin处理胃癌细胞后,DNA凝胶电泳结果显示呈明显的梯状条带;吖啶橙（AO）染色观察示细胞核内可见浓染致密的颗粒荧光、新月型改变、核固缩或片段化及凋亡小体的形成;流式细胞仪分析Emodin能浓度依赖性诱导MKN45细胞凋亡和G2/M期阻滞。结论：Emodin对人胃癌细胞MKN45有明显的生长抑制作用,且该作用与诱导细胞凋亡有关。

Research of Emodin on Proliferation of Human Gastric Cancer Cells MKN45 and Apoptosis

Objective：To study emodin on proliferation and induced apoptosis of human gastric cancer cell MKN45.Method：The growth inhibition of Emodin on MKN45 was observed by MTT;analysis of Emodin-induced apoptosis by DNA Ladder gel electrophoresis,fluorescence staining and flow cytometry.Result：Compared with control group,Emodin significantly inhibited the growth of MKN45 cells in a dose and time dependent,the IC50（48 hours） （47.5±0.3）μmol/L;after treatment of Emodin on gastric cancer cells,DNA gel electrophoresis results showed a clear laddering;acridine orange（AO） staining showed the nucleus visible fluorescent stain dense granules,crescent change, nuclear condensation or fragmentation and apoptotic body formation,flow cytometry analysis of Emodin concentration-dependent induction of apoptosis in MKN45 cells and G2/ M phase arrest.Conclusion：The growth of human gastric cancer cells MKN45 significantly was inhibited by Emodin,and the effect was related to apoptosis.
