绿茶多酚对D-半乳糖与Aβ25~35诱导的AD模型小鼠学习记忆障碍的改善作用

摘    要:目的:建立D-半乳糖与Aβ25~35诱导的AD模型小鼠,观察绿茶多酚对其学习记忆障碍的改善作用。方法:皮下注射D-半乳糖(120 mg/kg,连续给药12 w),并在第7 w时于侧脑室注射Aβ25~35(4 nmol/L),建立AD模型小鼠。利用跳台法、水迷宫、避暗法和开场实验评价绿茶多酚对AD模型小鼠学习记忆障碍的改善作用。结果:绿茶多酚明显延长跳台潜伏期,减少跳台错误次数;缩短游出迷宫时间和减少进入盲端的错误次数;减少避暗活动错误次数和延长进入穿梭箱潜伏期;同时,明显增加AD小鼠的探究行为。结论:绿茶多酚对D-半乳糖与Aβ25~35诱导的AD模型小鼠学习记忆障碍具有改善作用。

Abstract:OBJECTIVE: To study the effects of green tea polyphenol (GTP) on the behaviour of Alzheimer' s Disease like mice induced by D-galactose and Abeta25-35. METHODS: D-galactose (120 mg/kg) was intraperitoneally injected into mice for 12 weeks and Abeta25-35 (4nmol) was injected by icv to establish Alzheimer' s Disease like mice. The protective and therapeutic effects of GTP were determined by using water maze test, step down test, step through test and open field test. RESULTS: GTP ameliorated the deleterious effects of D-galactose and Abeta25-35, and thereby improved the animal' s learning and memory, prolonged latency time, and the error numbers were significantly reduced. At the same time the autonomic activities were significantly increased. CONCLUSION: GTP can improve the behaviour of Alzheimer' s Disease like mice induced by D-galactose and Abeta25-35.
