寒凝证动物模型的建立

摘    要:目的:探讨建立寒凝证动物模型的最佳温度条件。方法:将50只雌性SD大鼠随机分为A、B、C、D、E 5组,每组10只,A组为对照组,B、C组为-15℃冷冻组,D、E组为-25℃冷冻组,各组均冷冻5 d,每天冷冻4 h。C、E组同样冷冻后,正常喂养5 d。观察大鼠体征和体重,在冷冻结束后第1天和第5天取血检测血液流变学指标。结果:与A组比较,B、C、D、E组大鼠体征变化评分均明显升高,P<0.05;与A组比较,B组和D组冷冻期内大鼠体重下降,P<0.05,E组冷冻结束后体重增加较少,P<0.05;与A组比较,B组与D组全血还原黏度高切明显升高,P<0.05,红细胞刚性指数有升高趋势,但未见统计学差异。结论:每天-25℃、4 h,连续冷冻5 d,可建立寒凝证模型,结合整体体征评分及血液流变学指标评价,符合中医寒凝证候特点。

Establishment of animal model with cold coagulation syndrome

Abstract:Objective:To explore the optimum temperature conditions of animal model with cold coagulation syndrome.Methods: Fifty female SD rats were randomized into A,B,C,D and E groups,with 10 cases in each.A group was control group,B and D groups were-15 ℃ frozen groups,C and E groups were-25 ℃ frozen groups.Each group was put in deep freezer 5 hours every day,continous 5 days.D and E groups were fed 5 days.To observe symptoms,weight and hemorheology of rats at the first and fifth days after frozen days.Results:Compared with A group,outlooking scores of B,C,D,E groups increased remarkably(P< 0.05).Compared with A group,weight of B,D groups in frozing period reduced significantly(P< 0.05).Frozen after,weight of E group less increase(P< 0.05).Compared with A group,whole blood reductive viscosity high shear of B and D groups were significantly higher(P< 0.05),the erythrocyte rigidity indexes were increased,but there was no significant differences.Conclusion:cold coagulation syndrome models can be established as follow:-25 ℃,4 h,continuous freezer 5 d.Based on the overall symptoms score and hemorheology evaluation,the model has the characteristics of TCM cold coagulation syndrome.
