旋转培养仪在细胞培养中的应用

摘    要:目的:探讨旋转细胞培养仪(rotary cell culture system,RCCS)在组织工程学中的应用及组织工程学研究中种子细胞的扩增方法问题.方法:将多孔羟基磷灰石(porous hydroxyapatite,PHA)与骨髓基质细胞(bone marrow stromal cells,BMSC)在旋转培养仪中进行旋转复合培养,电镜(scan electric microcopy,SEM)观察细胞在PHA上的生长和增殖情况,与培养板培养法对比.并行AKP染色.结果:普通培养板中细胞主要黏附在PHA与培养板相接触的一侧,在载体的内部难以见到细胞分布.试验组中细胞均匀分布于材料的各个部位,细胞突起相连,交织呈网状.试验组中细胞数量多于对照组.AKP染色与旋转培养前无变化.结论:应用旋转细胞培养仪进行三维立体培养有利于细胞在支架上生长,提高细胞数量,改善细胞状况.对细胞分化无影响.

Abstract:Objective:To find out how the application of the RCCS and how to increase cell quantity and improve cell status in tissue engineering.Methods:Complex of cells and PHA cultured for 8 hours on common culture plate were transferred into RCCS to rotarial cultivate,observed by SEM and counted cells after digestion by trypsin.Results:Cells in common culture plate were mainly adhere at the side contacted to culture plate and few in the PHA.Cells in RCCS distributed at each department of PHA and had many pseudopodia contacted each other to form like a network.The numker of cells in rccs are morethan in conrnon cuttare plate.The AKP dyeing has no changl.Conclusion:The RCCS can accelerate cell's growth,increase cell's quantity and improve cell's status.
