复方芪苓配方颗粒对大鼠高尿酸血症的调控作用

摘    要:目的探讨复方芪苓配方颗粒治疗高尿酸血症大鼠的作用机制。方法通过腺嘌呤灌胃及氧嗪酸钾皮下注射建立高尿酸血症模型,连续给药14 d。分别测定血清尿酸(UA)、肌酐(Scr)、尿素(Urea)及胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL-c)和低密度脂蛋白(LDL-c)水平;采用RT-PCR方法分别测定大鼠肾脏中相关转运体的表达水平。结果与模型对照组比较,复方芪苓配方颗粒能显著降低高尿酸血症大鼠血清UA、Scr、Urea、TC、LDL-c水平,同时可以显著下调肾脏尿酸盐转运体(URAT1)mRNA的表达,上调有机阴离子转运体1(OAT1)、有机阳离子转运体2(OCT2)mRNA的表达。结论复方芪苓配方颗粒能改善高尿酸血症大鼠的肾功能、血脂代谢,其作用机制可能是下调大鼠肾脏中URAT1 mRNA的水平,上调OAT1和OCT2 mRNA的水平,从而降低肾脏尿酸盐重吸收能力及增加尿酸盐的分泌。

Regulatory effect of compound Qiling formula granules on hyperuricemia in rats
Abstract:Objective To explore the mechanism of action of compound Qiling formula granules in treating rats with hyperuricemia.Methods The hyperuricemia model was established in rats by adenine gavage and hypodermic injection of oxonic acid potassium salt.The drug was administered continuously for 14 days.The levels of serum uric acid(UA),creatinine(Scr),Urea,total cholesterol(TC),triglyceride(TG),high-density lipoprotein-cholesterol(HDL-c),and low-density lipoprotein-cholesterol(LDL-c) were tested respectively.RT-PCR was used to measure the expression levels of related renal transporters of the subjects.Results Compared with the model control group,the levels of serum UA,Scr,Urea,TC,LDL-c of rats with hyperuricemia were significantly lowered by applying compound Qiling formula granules.The expression level of the mRNA of the urate reabsorption transporter-1(URAT1) was down-regulated significantly,and the expression levels of the mRNA of organic anion transporter-1(OAT1) and organic cation transporter-2(OCT2)were up-regulated significantly.Conclusion Compound Qiling formula granule can improve the renal function and lipid metabolism of rats with hyperuricemia.Its mechanism may be related to the down-regulation of the level of URAT1 mRNA,and the up-regulation of the levels of OAT1 and OCT2 mRNA in the kidney of the rats,both of which can reduce the reabsorption capacity of urate in the kidney and increase the secretion of urate.
