胚胎脊髓移植联合甲基强的松龙治疗脊髓损伤的实验研究

摘    要:目的采用胚胎脊髓移植及大剂量甲基强的松龙联合应用治疗脊髓损伤,观察两者有无协同作用.方法雄性SD大鼠50只,随机分为5组,A、B、C、D组为T12急性脊髓右侧半横断损伤后治疗组,A组给大剂量甲基强的松龙及胚胎脊髓移植联合治疗;B组行大剂量甲基强的松龙治疗;C组行胚胎脊髓移植治疗;D组为单纯损伤;E组为空白对照.治疗后24 h及8周后观察大鼠一般行为及运动功能变化,评分分析.病理观察包括治疗后8周神经纤维记数对比及辣根过氧化物酶示踪观察.结果病理学形态观察示A组损伤区横断面神经纤维记数较B、C、D组明显增多(P<0.01);A、C组在损伤区注入的辣根过氧化物酶可扩散至损伤区远、近端各约1.0 cm处.除A组左下肢感觉有部分恢复外,无明显行为学改变.结论大剂量甲基强的松龙及胚胎脊髓移植可协同修复损伤脊髓.

Abstract:Objective The combined treatment of fetal spinal cord transplantation(FST) and methyl-prednisolone(MP) was applicated as spinal cord injury therapy, in order to find out whether or not there was any synergistic action existed. Methods 50 male SD rats were divided randomly into 5 groups. In group A to D, the acute semi-sectional lesion attacked at the right half of T12 spinal cord of each rat. Group A was treated with the combination of large dose MP and FST;Group B were treated with large dose MP only; Group C with FST;Group D without any therapy and Group E was a blank control. The fetal spinal cord was achieved from the female rats with pregnancy of 14 days. The changes of general behavior and the motive function of the rats were observed,24 hours to 8 weeks after treatment, with evaluation and analysis. The pathological observation, including the accounting numbers of nerve fibers at the cross section of injured spinal cord retrode tracer of horseradish peroxidase(HRP). Results The nerve fibres in group A were much more than that of B,C and D at injured spinal cord cross sections(P<0.05). In the group A and C,the injected HRP could diffuse around 1.0 cm at both sides of injured part. No obvious behavior changes were found except partial sensory recovery in group A. Conclusion There was synergistic effects of the treatment on spimal cord injuries with the combination of large dose of MP and FST.
